福尔马林灭活HRSV疫苗(formalin-inactivated HRSV vaccine, FI-HRSV)临床试验的失败表明HRSV疫苗研发存在多重难以克服的技术困难,此后该疫苗的研发进展极其缓慢。近年来,HRSV蛋白结构的深入解析为HRSV疫苗研发提供了新靶点,使HRSV疫苗的研发策略迅速增多,许多HRSV疫苗相继进入关键性研究阶段。目前,全球有19种HRSV疫苗正在开展临床试验,还有许多不同类型的候选疫苗也正在进行临床前的测试。

HRSV属于肺炎病毒科，正肺病毒属，有包膜，是非节段性、单股负链RNA病毒，病毒形状呈圆形和丝状体2种，外膜有突起，大小为80 nm至几μm。HRSV只有1个血清型，根据抗原性分为两大亚型：A型和B型，不同亚型的主要结构蛋白有明显差异。HRSV无血凝素和神经氨酸酶，未发现溶血活性。HRSV很不稳定，乙醚、氯仿、0.25%胰酶、0.1%去氧胆酸钠等都能将其灭活。对热敏感，55 ℃、5 min可使90%病毒灭活。在pH≤3的酸性环境中很快被灭活，pH>4以上相对稳定。
