ATTO 647 is rhodamine-based fluorescent label for the red spectral region that has similar spectral properties to Cy5. It has strong absorption, high fluorescence quantum yield, high thermal and photostability, and its absorption and fluorescence are independent of pH from 2 to 11. ATTO 647 is suitable for single-molecule detection applications, high-resolution microscopy (such as SIM, STED), flow cytometry (FACS) and fluorescence in-situ hybridization (FISH). ATTO 647N NHS ester is used for labeling amino-containing molecules such as proteins, peptides and amino-modified oligonucleotides.

An NHS ester allows the functionalization of an amino moiety in a variety of molecules, including DNA and RNA oligonucleotides, as well as peptides or proteins. NHS esters are especially useful in situations where the equivalent phosphoramidites exhibit instability to the conditions of oligonucleotide synthesis and/or deprotection.
