FITC标记葡聚糖是由荧光素异硫氰酸酯(5-异构体)与相应的葡聚糖偶联合成的。FITC标记葡聚糖是黄色至橙色粉末,易溶于水或盐溶液,呈黄色溶液。该产品也可以溶于DMSO,甲酰胺和其它极性有机溶剂,但在低级脂肪醇,丙酮,氯仿和DMF中基本不溶。葡聚糖是来源于肠系膜白串珠菌B-512F的分支多糖,本质上是一种α-(1-6)链接的线性葡萄糖链。分支比例大约是5%。荧光素是通过稳定的硫代氨甲酰键链接的,标记过程不会引起任何葡聚糖的解聚。FITC标记葡聚糖,每个葡萄糖单元对应0.002到0.008 mol的FITC。 这些低取代度使葡聚糖电荷极小,这也是渗透性研究最基本的要求。

 

葡聚糖从细菌( 如白念珠菌) 代谢产物中提取的一种水溶性多糖,由D-葡萄糖以1→6键连接,并在支链点1→2、1→3和1→4处连接配糖而组成。根据来源不同,可得到平均相对分子质量不同的各种支链葡聚糖。最初,葡聚糖被用作血浆替代品,由于其具有生物可降解、非特异性细胞黏附、抵御蛋白吸附、价格便宜及易于结构改造等特点,成为载体材料研究的热点。


Dextran is a water-soluble polysaccharide extracted from the metabolites of bacteria (e.g. Candida albicans). It is composed of D-glucose linked by 1 → 6 bonds and linked by glycosylation at branch points 1 → 2, 1 → 3 and 1 → 4. According to different sources, various branched glucans with different average relative molecular weights can be obtained. At first, glucan was used as a substitute for plasma. Because of its biodegradability, non-specific cell adhesion, resistance to protein adsorption, low price and easy structural modification, it has become a hotspot in the research of carrier materials.
