巯基官能化PEG是一种单官能PEG衍生物可以用来修饰蛋白质、多肽以及其它材料或者小分子和生物材料。具有无反应性的巯基、半胱氨酸,马来酰亚胺和巯基在PH6.5-7.5很容易形成稳定的硫醚键。硫醇基团对金表面具有很高的亲和性,用于金纳米粒子或金膜修饰。聚乙二醇修饰后可提高溶解度和稳定性,降低蛋白质和肽的免疫原性,它也还能能抑制带电分子在修饰表面的非特异性结合。通过S-S二硫键轻松空气氧化形成二聚体,可通过还原剂逆转。对于可逆聚乙二醇化是有用的。
Thiol functionalized peg is a monofunctional peg derivative that can be used to modify proteins, polypeptides and other materials or small molecules and biomaterials. Non reactive sulfhydryl, cysteine, maleimide and sulfhydryl groups can easily form stable thioether bonds at pH 6.5-7.5. Thiol groups have high affinity for gold surface and are used to modify gold nanoparticles or gold films. PEG modification can improve the solubility and stability, reduce the immunogenicity of proteins and peptides, and inhibit the non-specific binding of charged molecules on the modified surface. Dimer is easily formed by air oxidation through S-S disulfide bond, which can be reversed by reducing agent. Useful for reversible PEGylation.
