RSV是一种反式单链RNA病毒,共编码11种蛋白,其中G和F表面蛋白在RSV吸附和融合过程中发挥重要作用,是产生中和抗体最重要的抗原位点,也是诱导机体产生免疫原性和抗病毒的最主要靶点。

分别构建携带RSV-B1株F基因截短片段和全长的重组腺病毒rAd-F0DeltaTM和rAd-F0。在BALB/c小鼠中进行的免疫原性比较研究表明,每种载体均能诱导产生与RSV-long和RSV-A2病毒交叉反应的rsv - b1特异性抗体。抗rsv - b1抗体具有中和活性,对rsv -长株和RSV-A2分离株也具有较强的交叉中和活性。细胞免疫反应分析显示,rAd-F0DeltaTM和rAd-F0均能诱导Th1和Th2表型的CD4(+) t细胞应答和F蛋白特异性ctl。rAd-F0DeltaTM和rAd-F0免疫小鼠脾细胞产生的Th2细胞因子(IL-4、IL-5和IL-13)明显低于热灭活RSV-B1 (hirv - b1)免疫小鼠。总体体液和细胞免疫应答的比较表明,rAd-F0DeltaTM较rAd-F0具有更强的免疫原性。与接种hirv - b1的小鼠相比,两种重组腺病毒载体产生的抗病毒免疫对RSV-B1活病毒攻击产生了保护作用,原因是肺内恢复的病毒载量较低,体重减轻的恢复速度较快,嗜酸性粒细胞计数较低。这些研究结果表明,rAd-F0DeltaTM或rAd-F0具有符合RSV候选疫苗预期要求的免疫原性。
